| Abstract: | Prx6 attracts particular attention because of its ability to reduce the widest range of peroxidative substrates among the peroxiredoxin (Prx) family. In this study, TcPrx6a and TcPrx6b were cloned from Tribolium castaneum. RT-qPCR analysis revealed that TcPrx6a and TcPrx6b were highly expressed in fat body and head, respectively, and both were upregulated after exposure of the beetles to paraquat and deltamethrin. Knockdown of TcPrx6a and TcPrx6b decreased the tolerance of T. castaneum to paraquat-induced oxidative stress and deltamethrin, and enhanced deltamethrin-induced accumulation of H 2 O 2. Interestingly, knockdown of TcPrx6a and TcPrx6b negatively affected the pupation and eclosion rates as well as fecundity and fertility of beetles. Additionally, both RNAi and the dual-luciferase reporter assay in Sf9 cells revealed that the transcription of TcPrx6a and TcPrx6b was potentially regulated by transcription factor CncC. These results provided insights into the function of TcPrx6a and TcPrx6b as well as the transcriptional regulation of insect Prx6s.
