Abstract: Due to the retained cell membrane components, neutrophil membrane (Ntm) coatings can escape immune recognition by camouflaging and being accumulated in the inflammation site to have longer circulation times and nonimmunogenicity. The conventional method to obtain Ntm using freeze–thaw cycles is time-consuming and lacks efficiency. A polarization method has been developed to generate double layer forces on neutrophils to produce neutrophil membranes (Ntm) within 15 s under alternating electric fields (AEF) at 200 kHz and 3 V. The Ntm obtained by the polarization method and nylon-6 (Ny6) with 5-fluorouracil (5Fu) were employed as sheath and core, respectively, to prepare core/sheath fibrous mats using coaxial electrospinning for the capture of circulating tumor cells (CTCs), which were sequentially modified with streptavidin and anti-EpCAM. A low dosage (0.6 wt%) of 5Fu within the Ny6 reduces the outspread pseudopodia area and enhances the capture capacity of CTCs. The fibrous mat was adhered within a homemade microfluidic chip to investigate the capture capacities of CTCs. The maximum adsorption capacity of CTCs on the Ny6(5Fu)/Ntm is approximately 41 cells/mm2, about 2-fold higher than that on the Ny6/Ntm (21 cells/mm2). The outer Ntm coating not only effectively enhances the non-biofouling properties and cell viability but also significantly decreases hemolysis. In spiked blood samples, our Ny6(5Fu)/Ntm exhibited 95.7 % of the separation efficiency in a continuous system at a flow rate of 4 mL/hour with high selectivity. Our proposed method provides an ideal fibrous mat to pack within a microfluidic chip to capture CTC for precision medicine in cancer treatment as well as blood purification. [ABSTRACT FROM AUTHOR]
