Abstract: The Jintan Salt Mine in China has a long history and substantial reserves, with a salinity level reaching 85.9%. In order to gain a deeper understanding of the microbial community structure and its functional properties in the Jintan Salt Mine, and to assess its potential impact on the salt cavern hydrogen storage project, further investigation is required. The diversity of microbial community structure and function in the brine of the Jintan Salt Mine, which contains up to 30% salinity, was assessed using culture and macrogenomic approaches. A total of 71 bacterial strains and 5 archaeal strains were isolated and purified from brine using different media. After 16S rRNA molecular characterization, the isolated 71 strains were identified as members of Marinobacter, Halomonas, Halovibrio, Sutcliffiella, Bacillus, Halobacillus, and other genera. Furthermore, the metagenomics method revealed that the dominant phyla in brine were Euryarchaea, Proteobacteria, Balneolaeota, and Bacteroides. Metabolism was also found tobe the main functional activity of microorganisms in the brines. The metabolic processes of sulfur, methane, and carbon fixation in prokaryotes represent significant forms of energy metabolism. These processes are attributed to the presence of methanogenic and sulfate-reducing bacteria in brine samples. The findings of this study will considerably enhance the extant knowledge of microorganisms in salt mines. [ABSTRACT FROM AUTHOR]
