Oxidative stress (OS) has been extensively investigated for cancer treatment. However, existing reactive oxygen species (ROS)-based therapies are often compromised by the tumor antioxidant system and lack efficient strategies to amplify OS. Here, we developed a cascade-amplified nanoplatform (COP-Mn NPs) by assembling with COTIC-4F, camptothecin and manganese carbonyl (Mn 2 (CO) 10) that integrates photodynamic therapy (PDT), photothermal therapy (PTT), chemotherapy, chemodynamic therapy (CDT), and gas therapy to synergistically disrupt the antioxidant defense and trigger explosive ROS generation. In COP-Mn NPs, COTIC-4F generates ROS and heat for PDT and PTT, respectively. Camptothecin not only directly induces tumor cell death via chemotherapy but also enhances intratumoural hydrogen peroxide (H 2 O 2) concentration through nicotinamide adenine dinucleotide phosphate oxidase activation. The photothermal effect further drives H₂O₂ and Mn₂(CO)₁₀ to produce hydroxyl radicals and CO, enabling CDT and gas therapy. This multi-mechanistic cascade induces a sustained ROS storm, overcomes tumor antioxidant program, and significantly enhances immunogenic cell death compared to single-modal ROS therapies. Moreover, NIR-II fluorescence imaging guides precise tumor positioning. Our work provides a paradigm for designing OS-amplified nanoplatforms with improved efficacy against tumors.
