| Abstract: | Low-temperature and dehydration during freeze-drying severely impaired the viability of Lactiplantibacillus plantarum. This study evaluated the protective effects and underlying mechanisms of a novel cryoprotectant combining natural deep eutectic solvents (NADESs) with skim milk. Systematic comparisons were made among groups treated with NADESs alone (N), skim milk alone (M), or their composite (NM). Analyses included viability assays, gene expression, membrane fatty acid composition, and cell wall integrity. The NM group showed the highest survival rate (86.26 ± 2.02%). The composite's efficacy was attributed to a multi-level synergistic mechanism: physically, a dense NADES-protein matrix inhibited ice-crystal damage, molecularly, NADESs likely stabilized milk proteins and membrane integrity, metabolically, skim milk nutrients drove key stress-response pathways—methionine-quorum sensing, fatty acid metabolism, and active cell wall synthesis. This work first reveals a "methionine metabolism–quorum sensing–fatty acid metabolism–cell wall synthesis" synergistic network, providing a theoretical basis for designing advanced microbial cryoprotectants.
