| Abstract: | A combined theoretical–experimentalstudy of the hyperchromiceffect as occurring in the denaturation of a double stranded polyA-polyTis presented. Our theoretical/computational procedure allows us toreproduce the essential features of the experimental spectra and tocharacterize those molecular interactions responsible for the changesin the UV absorbance. We found that although excitonic intrastrandinteractions strongly affect the absorbance, they are almost fullymaintained in the single-stranded DNA. Our data indicate that hyperchromiceffect originates from the higher delocalization of the excitonicstates in the denaturated DNA with respect to the double-strandedconformation.
