Abstract (English): Introduction. Exposure to organic solvents (OS) and paints has been associated with genotoxicity and increased risk of neoplasms. However, the type of DNA damage induced for this exposure has not been yet fully characterized in humans. This knowledge would explain the mechanisms of genotoxicity caused by these compounds. Car painters in informal sector are a group highly exposed to OS and paints, due to inadequate practices of occupational safety. Objective. To identify the oxidative and methylating damage in the DNA of lymphocytes of car painters exposed to OS and paints. Materials and methods. Isolated peripheral blood lymphocytes from 62 painters and 62 unexposed subjects were analyzed by the high-throughput comet assay, coupled to the Fpg and AlkA enzymes. The categories of DNA damage evaluated were basal damage (without enzymes), oxidative and methylating damage. The parameter used to measure the damage was the percentage of DNA in tail (%TDNA). Results The %TDNA was higher in the exposed group compare to the unexposed (p < 0.05). In the exposed group, the %TDNA was higher in the oxidative damage category than the baseline (16.50 vs 12.87, p < 0.001), whereas the methylating damage did not show significant differences (14.00 vs 12.87; p > 0.05). Conclusion. In this study, exposure to OS and paints was associated with an increase in oxidative lesions in the DNA of car painters lymphocytes, such as 8- oxodG and other formamidopyrimidine products, which are considered highly mutagenic. Abstract (Spanish): Introducción. La exposición a solventes orgánicos (SO) y pinturas ha sido asociada con genotoxicidad y mayor riesgo de neoplasias. Sin embargo, aún no ha sido bien caracterizado el tipo de daño en el ADN que induce esta exposición en humanos. Este conocimiento permitiría aclarar los mecanismos de genotoxicidad originada por estos compuestos. Uno de los grupos con mayor exposición a SO y pinturas son los pintores de carros del sector informal, quienes trabajan sin adecuadas prácticas de seguridad ocupacional. Objetivo. Identificar el daño oxidativo y metilante en el ADN de linfocitos de pintores de carros expuestos a SO y pinturas. Materiales y métodos. Linfocitos aislados de sangre periférica de 62 pintores y 62 sujetos no expuestos fueron analizados mediante el ensayo cometa de alta eficiencia, acoplado a las enzimas Fpg y AlkA. Las categorías de daño en el ADN evaluadas fueron daño basal (sin enzimas), oxidativo y metilante. El parámetro de medición del daño fue porcentaje de ADN en cola (%ADNC). Resultados. El %ADNC fue mayor en el grupo expuesto con respecto al no expuesto (p <0,05). En el grupo expuesto el %ADNC fue mayor en la categoría daño oxidativo con respecto al basal (16,50 vs 12,87; p <0,001), mientras que el daño metilante no mostró diferencias significativas (14,00 vs 12,87; p >0,05). Conclusión. La exposición a SO y pinturas se asoció con un aumento de lesiones oxidativas en el ADN de linfocitos de pintores de carros, tales como 8-oxodG y otros productos formamidopirimidina, los cuales son considerados altamente mutagénicos.
