Abstract (English): Justicia pectoralis Jacq., widely used in traditional Cuban medicine as sedative and anxiolytic. Its main components, coumarins, have a recognized sedative activity, are used as markers in various extracts obtained from this plant. The hydroalcoholic extract 30% is used as raw material in the preparation of syrup. Microwave assisted extraction is of recognized importance because of the speed and efficiency in obtaining phytoconstituents compared to conventional methods and Response Surface Methodology is an efficient statistical tool that has been successfully used in testing process parameters and their interactive effects. The objtetive of this work was optimize the obtaining of the hydroalcoholic extract 30% of Justicia pectoralis Jacq. by means of microwave energy through a Box-Behnken design under the response surface methodology. A Sanyo EM-T109SS modified domestic microwave oven was used. Three variables irradiation power (540 – 900 W), time (2-5 min), and liquid to solid ratio (10 -20 mL/g) were regarded as factors in the response surface design of Box-Behnken of 15 executions, including 3 center points. The yield of total coumarins was used as the response variable. The optimal conditions were 4 min at 540 W and a liquid to solid ratio of 20 ml/g for a predicted value of total coumarins of 0.48 mg/g. Compared to the traditional method by reflux (2 h and 0.42 mg/g), the EAM demonstrated significant time savings, with greater recovery of coumarins. The results showed that microwave-assisted extraction can be used as an efficient method to obtain hydroalcoholic extract of Justicia pectoralis Jacq. [ABSTRACT FROM AUTHOR]
Abstract (Spanish): Justicia pectoralis Jacq. es ampliamente utilizada en la medicina tradicional cubana como sedante y ansiolítico. Sus componentes principales, las coumarinas, de una reconocida actividad sedante, se emplean como marcadores en diversos extractos obtenidos a partir de esta planta. El extracto hidroalcohólico al 30% se emplea como materia prima en la elaboración de un jarabe. La extracción asistida por microondas es de reconocida importancia por la rapidez y la eficiencia en la obtención de fito constituyentes, comparado con los métodos convencionales y la metodología de superficie respuesta es una herramienta estadística utilizada de manera exitosa para la evaluación de los parámetros de extracción y sus interacciones. El objetivo del trabajo fue optimizar la obtención del extracto hidroalcohólico al 30% de Justicia pectoralis Jacq. mediante la energía de las microondas a través de un diseño Box-Behnken bajo la Metodología de superficie respuesta. Tres variables: potencia de irradiación (540 – 900 W), tiempo (2 – 5 min) y relación disolvente material vegetal (10 – 20 mL/g) fueron consideradas como factores en el diseño de superficie respuesta Box-Behnken de 15 corridas incluyendo 3 puntos centrales. El rendimiento de coumarinas fue utilizado como variable respuesta. Las condiciones óptimas fueron 4 min de irradiación a 540 W y una relación disolvente/ material vegetal de 20 mL/g para un valor predicho de coumarinas totales de 0.48 %. Frente al método tradicional por reflujo (2 h y 0.42 mg/g % de rendimiento) la extracción asistida por microondas demostró significativos ahorros de tiempo con mayor recobrado de coumarinas. Los resultados mostraron que la extracción asistida por microondas puede ser empleada como un método eficiente para la obtención de extracto hidroalcohólico de Justicia pectoralis Jacq.
