Abstract (English): INTRODUCTION: Oxidative stress plays a significant role in tumor progression and the response to treatment. Antioxidant enzymes, including glutathione peroxidase (GPx) and glutathione reductase (GR), help protect cells from oxidative damage. Changes in the activity of these enzymes may be linked to clinical outcomes in pediatric patients with sarcomas. OBJECTIVE: This study compared levels of GPx and GR in children with sarcomas to a control group without sarcomas. METHODOLOGY: An analytical observational study involved 30 children diagnosed with sarcomas and 43 children without sarcomas. All participants provided blood samples for plasma analysis, and a second sample was collected at the end of treatment for the sarcoma group. The concentrations of GPx and GR were measured using commercial kits from Enzo and BioAssay Systems. For the statistical analysis, medians, interquartile ranges, and the Mann-Whitney U test were utilized. RESULTS: Initial levels of GPx were found to be higher in children diagnosed with sarcomas, with a median of 164.4 U/mL ([Q25 114.9–Q75 188.85]), compared to the control group, which had a median of 108.83 U/mL (Q25 93.96–Q75 137.77; p < 0.05). After treatment, the final levels of GR were lower in the children with sarcomas, measuring 3.79 U/L (Q25 3.14–Q75 5.04), in contrast to the control group, which showed levels of 4.77 U/L (Q25 3.82–Q75 5.45; p < 0.05). CONCLUSIONS: Sarcomas and their treatment can induce changes in cellular redox status, particularly by initially activating GPx and decreasing the final activity of GR. These alterations could influence the prognosis of pediatric patients with sarcomas. [ABSTRACT FROM AUTHOR]
Abstract (Spanish): INTRODUCCIÓN: El estrés oxidativo está implicado en la progresión tumoral y respuesta al tratamiento. Las enzimas antioxidantes, como glutatión peroxidasa (GPx) y glutatión reductasa (GR), defienden a la célula de este daño oxidativo. En pacientes pediátricos con sarcomas, su actividad podría estar alterada y asociarse a la evolución clínica. OBJETIVO: Comparar los niveles de GPx y GR entre niños con sarcomas y un grupo control sin sarcomas. METODOLOGÍA: Estudio observacional analítico con 30 niños con sarcomas y 43 sin sarcomas. A todos se les obtuvo una muestra sanguínea para análisis plasmático; en los pacientes con sarcomas se tomó una segunda muestra al finalizar el tratamiento. Las concentraciones de GPx y GR se determinaron mediante kits comerciales (Enzo y BioAssay Systems). Se utilizaron medianas, rangos intercuartílicos y U de MannWhitney para análisis estadístico. RESULTADOS: Los niveles iniciales de GPx fueron mayores en niños con sarcomas (mediana 164.4 U/mL, Q25 114.9–Q75 188.85) que en el grupo control (108.83 U/ mL, Q25 93.96–Q75 137.77; p < 0.05). Después del tratamiento, los niveles finales de GR fueron menores en los niños con sarcomas, midiendo 3,79 U/L (Q25 3,14–Q75 5,04), en contraste con el grupo control, que mostró niveles de 4,77 U/L (Q25 3,82– Q75 5,45; p < 0,05). CONCLUSIONES: Los sarcomas y su tratamiento podrían inducir alteraciones en el estado redox celular, particularmente una activación inicial de GPx y disminución de la actividad final de GR, lo cual podría influir en el pronóstico de los pacientes pediátricos con sarcomas.
