Abstract (English): The prevalence of diabetes in Mexico has increased considerably over the past 20 years, thus increasing the need for the development of non-invasive and reliable glucose monitoring platforms. The present research focus on the development and evaluation of a novel non-invasive glucose monitor for the quantification of glucose levels in sweat. This platform is based on the use of screen-printed electrodes modified with a Prussian Blue (PB) electrochemical mediator and the glucose oxidase (GOx) enzyme to provide glucose specificity. Additionally, we present the design of a portable electronic device for generating the input signal and processing the analyte output signals. The system was evaluated in a 0.1 M phosphate buffered saline (PBS) solution supplemented with different physiologically relevant glucose concentrations. The results demonstrated that the PB mediator plays a key role in the detection of glucose, since the control electrode modified with the enzyme but lacking the PB mediator did not show trend, while the electrode modified with PB and enzyme showed a good correlation coefficient with analytical sensitivity of 0.0008 mA µM-1. This finding could revolutionize the field of glucose monitoring, facilitating more accessible and less invasive health technologies. [ABSTRACT FROM AUTHOR]
Abstract (Spanish): La prevalencia de diabetes en México ha aumentado considerablemente en los últimos 20 años, aumentando así la necesidad de desarrollar plataformas de monitoreo de glucosa no invasivas y confiables. La presente investigación se enfoca en el desarrollo y la evaluación de una novedosa plataforma de monitoreo no invasiva para la cuantificación de niveles de glucosa en el sudor. Dicha plataforma se basa en el uso de electrodos impresos modificados con un mediador electroquímico de Azul de Prusia (PB) y con una enzima altamente específica, como la glucosa-oxidasa (GOx). Adicionalmente, presentamos el diseño de un dispositivo electrónico portátil para la generación de la señal de perturbación y el procesamiento de las señales de respuesta del analito. El sistema se evaluó en una solución buffer de fosfato salino (PBS) 0.1 M, suministrada con diferentes concentraciones de glucosa adecuadas fisiológicamente. Los resultados demostraron que el mediador de PB desempeña un papel fundamental en la detección de glucosa, debido a que el electrodo modificado con la enzima, pero sin el mediador PB, no mostró tendencia, mientras que el electrodo modificado con PB y enzima presentó un buen coeficiente de correlación, con una sensibilidad analítica de 0.0008 mA µM-1.
