| Abstract (English): | The article focuses on the role of the AGO4 gene in rice resistance to the rice hoja blanca virus (RHBV). Using CRISPR/Cas9 technology, mutations were produced in the AGO4 gene of the Fedearroz 2000 variety, resulting in 14 edited plants that showed increased susceptibility to the virus. Phenotypic evaluations and expression analysis of the AGO4 gene indicated that the edited lines exhibited profiles similar to those of a susceptible control. Additionally, modeling of the AGO4 protein structure revealed significant changes that support its function in resistance to RHBV. [Extracted from the article] |
| Abstract (Spanish): | Introducción: El virus de la hoja blanca del arroz (RHBV), transmitido por el insecto vector Tagosodes orizicolus, representa una amenaza significativa para el cultivo del arroz. Métodos: Utilizamos la tecnología CRISPR/Cas9 para producir mutaciones específicas en el gen AGO4 de Oryza sativa, utilizando la variedad Fedearroz 2000, con el objetivo de dilucidar la participación del gen en la resistencia a RHBV. Resultados: Obtuvimos 14 plantas editadas que presentaron deleciones de uno, dos y tres nucleótidos en la secuencia del exón 23 del gen AGO4. Las evaluaciones fenotípicas mostraron un aumento en la susceptibilidad a RHBV en las líneas editadas. Identificamos la presencia de RHBV en el tejido foliar de plantas infectadas mediante la amplificación de los genes de la nucleoproteína, NS3 y NS4 del virus. Mediante RT-qPCR analizamos los patrones de expresión del gen AGO4, lo cual mostró que, en las líneas editadas, los perfiles de expresión son similares al control susceptible. Además, el modelado de la estructura terciaria de la proteína AGO4 y su variante mutante demostró cambios en el dominio PIWI y la presencia de la tríada catalítica DDH, lo que confirma su papel en la mediación de la resistencia al RHBV.
