Abstract: The polysaccharide Vi has proven to be a protective antigen in young adults. In controlled clinical trials in areas with high rates of typhoid fever, the protective efficacy of the Vi was approximately 70%. However, methods were used, to synthesize polysaccharide Vi-protein conjugates in order to both enhance the antibody response and confer T-dependent properties to the polysaccharide in populations at high risk. In this study the immunogenicity and type of immune response (Th2, IgG1 or Th1, IgG2a) to a polysaccharide Vi-protein conjugates was evaluated in mice. The purified polysaccharide was linked to carrier proteins via carbodiímida-mediated reaction, with adipic acid dihydrazide as spacer arms. This synthetic scheme was reproducible, provided high yields of Vi-protein conjugates, and was applicable to several medically relevant proteins such as Outer Membrane Vesicle from Neisseria meningitidis serogrupo B and tetanus toxoids. The resultant conjugate induced mainly IgG2a subclass anti-Vi and was more immunogenic than the Vi alone in mice. In contrast to the T-independent properties of the Vi, conjugates of this polysaccharide with several medically relevant proteins induced booster responses and induce an increase the duration of their immune response in the time, in mice. The serum of animals inoculated with conjugate was shown high titers IgG anti-carrier proteins, like proteins native. [ABSTRACT FROM AUTHOR]
