The Institute of Testing of Materials of the Engineering School of Montevideo developed a standard method in reference to the quality of biological tissues of human origin currently employed in clinical human transplantation in our country. In a joint study with researchers from the Medicine (BNOT) and the Chemistry Schools, the authors tested cardiovascular tissues preserved by programmed liquid nitrogen freezing, evaluating quality criteria. Comparative tests of molecular crystalographic and biomechanical variables in the fresh and defrosted condition were carried out in laboratory setting. Preliminary results are shown. The biomechanical assays were carried out on six fresh and seven defrozen anular segments, obtained from donors of BNOT. In relation to the donors selection, ablative and vessels dissection techniques, procedures regulated by BNOT were applied. With the same criteria tissues in the antibiotic sterile areas were processed , the use of cryopreservation methods, bottling, programmed freezing curve and storage in liquid nitrogen vapors to -142 ºC . Transversal uniaxial traction tests were made, as X rays diffraction tests from its internal aspects were also made, analysing crystalline structures on fresh and frozen samples. It is concluded for that sample, that although there is an increased stiffness in the defrosted stuff, it is not statistically significant. Instead, the x ray diffraction profiles, show modifications in the molecular arrangement between fresh and defrosted samples which allow to assess the thermal effect and so evaluate the quality of the procedure. The complex biomechanical behaviour of the vascular polymers and the effect provoked on them by the cryogenic conditions, need further multi-disciplinary studies in its functional and structural aspects, that this group of researchers continues to carry out. El Instituto de Ensayo de Materiales de la Facultad de Ingeniería desarrolló un modelo de investigación referido al análisis de calidad de biomateriales de origen humano, aplicado a la medicina del trasplante en Uruguay. Conjuntamente con las Facultades de Medicina (BNOT) y de Química se analizaron criterios de calidad para tejidos cardiovasculares conservados por congelamiento programado en nitrógeno líquido. Se hicieron estudios de valoración comparativos de variable biomecánicasas y cristalográfico moleculares entre el material fresco descongeladodo fuera de parámetros fisiológicos. Se muestran resultados preliminares. Los ensayobiomecánicosos se efectuaron sobre seis segmentos anulares frescos y sietdescongeladosos de aorta torácica descendente provenientes de donantes deBNOTOT. Se aplicaron procedimientos reglados por eBNOTOT en selección de donantes, técnicas de ablación y disección de vasos. Con iguales criterios se procesaron los tejidos en las áreas de esterilización antibiótica, uso de medios de criopreservación, envasado, curva de congelamiento programado y almacenamiento en vapores de nitrógeno líquido a -142 °C . Se practicaron ensayos de tracción uniaxial trasversa y análisis de difracción de rayos X por sus caras íntimas para materiales de estructuras cristalinas sobre las muestras frescas y congeladas. Se concluye para esa muestra que si bien resulta un aumento de rigidez en los descongelados, no es estadísticamente significativo. En cambio los perfiles difractográficos de rayos X muestran modificaciones en el ordenamiento molecular entre muestras frescas y descongeladas que permiten medir el efecto térmico y por lo tanto, evaluar la calidad del procedimiento. El complejo comportamiento biomecánico de los polímeros vasculares y el efecto que sobre él ejercen las condiciones criogénicas requieren de estudios multidisciplinarios en los aspectos funcionales y sus correlatos estructurales que el grupo continúa realizando.
