Abstract: This letter discusses the potential effect of the orexin receptor antagonist suvorexant on Parkinson's disease (PD) symptoms. Orexin is a neuropeptide involved in sleep regulation and other functions, and dysregulation of the orexin pathway is associated with the development and progression of PD. The letter suggests that suvorexant, which is approved for the management of sleep disorders, could be effective in treating sleep disorders and non-motor symptoms in PD patients. However, a case report showed that suvorexant use in a PD patient led to severe delirium and nightmares. The authors recommend further clinical trials to evaluate the clinical profile of suvorexant in PD and the efficacy of orexin receptor agonists in preventing PD progression.
