Abstract: The article offers the authors' insights on the benefits and defects of fragment-based screening (FBS) and diversity-oriented synthesis (DOS) methods in creating libraries of compounds for drug discovery. It says that FBS design offers varied libraries and improved use of synthetic resources. It tells that DOS libraries develop diverse molecules whose molecular weights are proximate to the masses of drug-like compounds. It adds that unlike DOS, FBS would not offer non-traditional target hints.
