Abstract: The article focuses on clarifying the role of the key characteristics (KCs) framework in evaluating carcinogenic mechanisms within the International Agency for Research on Cancer (IARC) Monographs. The KCs framework, developed from a review of established human carcinogens, provides a systematic and transparent method to assess mechanistic evidence without bias toward specific mechanisms. It emphasizes that individual KCs, such as "induces oxidative stress" or "is genotoxic," should not be used alone to establish strong mechanistic evidence or to classify agents as probable human carcinogens (Group 2A), but rather in combination with other evidence streams. The article also highlights that the KCs approach is increasingly applied by agencies like the US Environmental Protection Agency and the US National Toxicology Program to support cancer hazard identification and risk assessment.
