| Abstract: |
Quantity of secondary laticifers in vascular cambia of phloem tissue in the Hevea rubber tree is a principal factor for natural rubber output and is positively proportional to final latex yield in rubber production. Coronatine, a compound structurally similar to jasmonic acid, exhibits high activity for induction of secondary laticifer differentiation in the phloem tissues of Hevea brasiliensis , while its regulation mechanisms at protein level is unclear. Here, we performed comparative proteomics of vascular cambium tissues from one-year-old peripheral shoots of H. brasiliensis treated with coronatine, aiming to elucidate the molecular mechanism driving coronatine-induced secondary laticifer differentiation. Exogenous application of coronatine significantly promotes secondary laticifer differentiation in the bark of epicormic shoots of the H. brasiliensis. Overall, proteomic analysis identified 74 differentially accumulated protein spots through 2-D DIGE coupled with MALDI TOF/TOF MS method. Gene ontology enrichment analysis showed that the 74 differentially accumulated proteins were tightly associated with the S-adenosylmethionine metabolic process, cytoplasm process, and ATP binding process. Eight of them, especially heat shock proteins, 14–3–3 proteins and 26S proteasome, showed consistent changed patterns with the publicly transcriptome data. Our findings offer new insights into the proteomic regulation associated with coronatine-mediated secondary laticifer differentiation in H. brasiliensis , along with their corresponding genes, which can be targeted for future genetic enhancement strategies aimed at improving rubber yield.
