Tightly focused femtosecond laser radiation is a convenient tool for manipulating biological objects. Recently, the method of femtosecond laser-induced DNA disruption within living cells has been actively developed, particularly relevant for assisted reproductive technologies in humans. To further advance this approach, it is essential to study the products of DNA dissociation induced by multiphoton ionization. In this work, structural changes in DNA were investigated using circular dichroism and Raman spectroscopy on a model system—an aqueous DNA solution. Hoechst 33 342 staining for adenine–thymine-rich regions revealed a decrease in the fluorescence intensity of irradiated DNA. Raman peaks characteristic of DNA also decreased after irradiation, and circular dichroism spectra showed distortion. These findings confirm the impairment of DNA structure following 1033 nm femtosecond laser irradiation. [ABSTRACT FROM AUTHOR] |
