| Abstract: |
Polymersomes, self-assembled from amphiphilic block copolymers, are nanoscale vesicles with a hydrophilic core and a hydrophobic bilayer. This unique structure allows them to efficiently encapsulate both hydrophilic and hydrophobic drugs, enabling co-delivery and synergistic therapeutic effects. This review systematically summarized their preparation methods, drug loading strategies, and functionalization approaches, highlighting recent applications in tumor-targeted therapy, gene editing, and anti-infection treatment. Despite ongoing challenges in scalable production, quality control, and clinical translation, polymersomes represent a promising intelligent platform for nano drug delivery with significant potential to enhance the treatment efficacy of drugs and reduce their side effects. HIGHLIGHTS: Polymersomes encapsulate both hydrophilic/hydrophobic drugs for synergy. Progress in their preparation, loading, and functionalization is summarized.
